Opicapone Evaluated for Patient Characteristics, Treatment Patterns, and Safety/Tolerability in Individuals with Parkinson Disease

In the Opicapone Treatment Initiation Open-Label Study (OPTI-ON) study, participants treated with opicapone (Ongentys; Neurocrine Biosciences, San Diego, CA) for Parkinson disease (PD) "OFF" episodes were observed for patient characteristics, treatment patterns, and safety/tolerability.

The data describes the baseline characteristics of participants enrolled in the study. All patients were initiated on once-daily opicapone 50 mg. At baseline, patients utilized a variety of carbidopa/levodopa (CD/LD) regimens, including immediate-release CD/LD tablets (Sinemet) (79.1% of participants) and extended-release CD/LD capsules (Rytary) (22.2% of patients). 12.6% of participants used entacapone before enrollment and were switched to opicapone and 2.9% of participants were receiving Deep Brain Stimulation (DBS) at baseline. 
 
The study is an open-label, single-arm, multicenter, observational, prospective, 6-month study of participants (n=239) with PD experiencing “OFF” episodes treated with opicapone capsules as adjunctive treatment to carbidopa/levodopa (CD/LD). Participants had a mean PD duration of 8.6 years and a mean (±standard deviation) age of 67.7 (±9.0) years. Participants ranged from “mild” to “advanced” PD severity at baseline, as assessed using the clinician-based PD Status Questionnaire. 

Findings from OPTI-ON will further characterize the PD population treated with opicapone from the real-world, clinician- and patient-reported outcomes data. These findings show that opicapone was added to a variety of CD/LD formulations, including over 22% of participants utilizing extended-release CD/LD capsules (Rytary). While the study was conducted during the ongoing pandemic, most investigators elected to initiate participants on opicapone with an in-person visit.
