The Food and Drug Administration (FDA) has approved a new indication for onabotulinumtoxinA (Botox; Allergan, Madison, NJ)—treatment of upper limb spasticity in children, age 2 to 17 years. The decision came after a 6-month priority review, which istypically granted to therapies with potential to provide significant improvements in safety and effectiveness compared with current standard of care. Approval is based on data from two phase 3 studies (NCT01603602, NCT01603615) a 12-week double-blind placebo controlled safety and efficacy study and a 1-year open-label extension study....
A new study published in Neurology shows people with well-controlled epilepsy are at risk of SUDEP. Researchers found SUDEP occurred in individuals who had not had a recorded seizure in the previous 12 months and those who missed a dose of antiepileptic medicine or were sleep deprived. The estimated incidence of SUDEP is 1 in 1,000 people with epilepsy per year, but only 16 percent of family members had heard of SUDEP. In 93% of the cases (n = 237), death was not witnessed; 70% of SUDEP occurred during sleep. In only 37% of cases, people had taken their last prescribed dose of...
The positive allosteric modulator (PAM) of GABAA channels, CVL-865 (Cerevel, Boston, MA) has been shown in a phase 2 trial (NCT02564029) to have pronounced anticonvulsant activity in patients with photosensitive epilepsy, with efficacy similar to lorazepam (used as a positive control). What sets CVL-865 apart as a GABAergic PAM is that it has minimal activity at channels containing the α1 subunit. Modulation of the α1 subunit has anticonvulsant effects but also causes the sedative and cognitive effects and addictive properties seen with non-selective GABAergics, such...
